imageRecently, the pharmaceutical industry has been heavily investigating and discussing continuous manufacturing. Key to this approach is the continuous supply of a tableting formulation to a fast-running tablet press.
 
The most challenging step in tablet manufacturing is choosing the correct formulation composition. Achieving the optimal balance between the active pharmaceutical ingredient (API) and excipients such as diluents, binders, disintegrants, colorings, flavorings, sweetening agents, lubricants, and glidants, is critical. It ensures that the final drug product functions properly, is practical and convenient to handle, and facilitates patient compliance.

Q: How does compaction dwell time influence scale-up from an R&D tablet press to a production press?
 
A: Robert Sedlock, Natoli Scientific, says:
 
imageMoving a drug product from a small R&D tablet press to a production press during scale-up has been an ongoing challenge in the pharmaceutical industry for many decades. Formulations don't always transfer successfully to the larger production machines. Developing the product on a production press would eliminate the transfer challenges, but the amount of formulation required to operate such a machine is very high and not cost effective. While many factors influence tablet quality, problems during scale-up often stem from a difference in the compaction dwell time between the R&D press and production press.
